Dendritic cells use a specialized pathway called cross-presentation to activate CD8+ T by presenting peptides from exogenous protein antigens on major histocompatibility complex class I molecules. Considerable evidence suggests that internalized cross endocytic membranes access cytosolic proteasomes for processing. The mechanism of dislocation represents unsolved problem. Here we describe the development sensitive reporter substrate, an N-glycosylated variant Renilla luciferase fused Fc region human IgG1. is designed be enzymatically inactive when glycosylated, but active after asparagine aspartic acid conversion occurs upon deglycosylation enzyme N-glycanase-1. generation luminescence depends internalization, deglycosylation, AAA-ATPase VCP/p97, and chaperone HSP90. By incorporating cell epitope into fusion protein, demonstrate antigen cytosol rate-limiting step in cross-presentation. Currently, are using this novel probe determine efficiency entry nanoparticles co-encapsulate ddRLuc-Fc mRNA. It will provide us with important information about mRNA delivery those nanoparticles, potentially leading breakthrough vaccine development.
